BINDING OF THE ANTITUMOR DRUG ADRIAMYCIN TO DNA-HISTONE COMPLEXES

چکیده مقاله:
Isotherms of the binding of the anthracycIine antibiotic, adriamycin (adriblastin), to DNA histone complexes was studied by means of spectroscopic analysis. The results indicated that: (a) binding of adriamycin to histones reduced the interaction of histones with DNA, (b) binding of the drug to DNA did not change the binding affinity of histone to DNA and, (c) in the explored binding range of r<O.1 the binding of adriamycin to DNA-histone complex proved to be anticooperative with n values of 0.32 for the interaction of histone with DNA-drug and 0.26 for the binding of DNA to histone-drug complex. The results suggest the possible participation of his tones in the DNA-drug complex formation.

متن کاملTight binding of the antitumor drug ditercalinium to quadruplex DNA.
The structural selectivity of the DNA-binding antitumor drug ditercalinium was investigated by competition dialysis with a series of nineteen different DNA substrates. The 7H-pyridocarbazole dimer was found to bind to double-stranded DNA with a preference for GC-rich species but can in addition form stable complexes with triplex and quadruplex structures. The preferential interaction of the dru...
متن کاملStructural comparison of anticancer drug-DNA complexes: adriamycin and daunomycin.
The anticancer drugs adriamycin and daunomycin have each been crystallized with the DNA sequence d(CGATCG) and the three-dimensional structures of the complexes solved at 1.7- and 1.5-A resolution, respectively. These antitumor drugs have significantly different clinical properties, yet they differ chemically by only the additional hydroxyl at C14 of adriamycin. In these complexes the chromopho...
متن کاملBinding of phosphorylated histone H1 to DNA.
A chromatin associated protein kinase was used to add 3 moles of phosphate to seryl side chains of 1 mole of histone H1. The DNA binding properties of this in vitro phosphorylated H1 were compared with those of unmodified H1. Considerably more radioactive superhelical DNA was retained on nitrocellulose filters at 20mM-40mM NaCl by phosphorylated H1 than by unmodified H1. However, zone velocity ...
متن کاملNMR studies on the binding of antitumor drug nogalamycin to DNA hexamer d(CGTACG).
The interactions between a novel antitumor drug nogalamycin with the self-complementary DNA hexamer d(CGTACG) have been studied by 500 MHz two dimensional proton nuclear magnetic resonance spectroscopy. When two nogalamycins are mixed with the DNA hexamer duplex in a 2:1 ratio, a symmetrical complex is formed. All non-exchangeable proton resonances (except H5' & H5") of this complex have been a...
متن کاملFT- SERS Study of Adriamycin - DNA Intraction
FT-SERS (Fourier Transform Surface Enhanced Raman Scattering) of adriamycin and its complex with DNA is reported. It is shown that in agreement with previous Raman studies the interaction of adriamycin with DNA takes place through an intercalation mechanism. The presence of a new band at 731 cm-1 suggests that ring D of adriamycin is not involved in the intercalation process.
